This poster highlights the HEK-mediated adeno-associated virus (AAV) production process and the development of an open-source production platform incorporating a chemically defined serum-free medium. Initial attempts using an open-source CHO-medium resulted in cell aggregation, slow growth, and an inability to transfect and produce AAV. Therefore, we explored creating an HEK-friendly medium and process conducive to healthy cell growth, transfection, and vector production. Nutrient consumption rates were studied to elucidate metabolic activity associated with AAV production. This poster was #835 at ISCT 2024
