Optimize Your REBEL’s Performance with a Preventive Maintenance Plan
1. REBEL Autosampler Sample Needle
Routine PM replacement and alignment of the autosampler needle will reduce chances of leaks, clogs, and potential missed injections during analysis.
2. REBEL Autosampler Sample Syringe
The autosampler syringe is responsible for all the fluidic movement and precise sampling within the REBEL. Over time the seals within the syringe can wear resulting in leaks and lowered analysis precision of fluidic transfer/sample loads.
3. REBEL Gas System Inlet Filter*
Annual PM replacement of the air inlet carbon and HEPA filters is to ensure optimal performance for the gas handling system in the REBEL. If not replaced the filters become less efficient and could result in poor analysis performance and contamination of more expensive components

4. REBEL Waste System Filter*
A particulate filter included in the waste system within the REBEL is to prevent material from entering the membranes of the waste pump. Routine replacement prevents clogging and decreased waste removal performance.

In upstream bioprocessing, cell culture media development is a key area of focus, and monitoring of the media all the way through manufacturing is critical. KNOW WHAT'S IN YOUR MEDIA
Understanding which key nutrients are present in your cell culture media is critical to media screening and optimization efforts. In this experiment, nine commercial CHO media were measured using the REBEL in a media screening evaluation (Figure 1). The amino acid profile reveals high diversity in nutrient composition across the media panel. With the REBEL you can measure key nutrients such as amino acids in both fresh and spent media at-line in real time to gain deeper insight into your process and obtain high titer faster.

REBEL measures over 30 analytes including all amino acids, certain vitamins, and bio amines. Plotted here are a subset of amino acids including all essential ones and others in 9 different CHO media. This comparison shows that the concentration of several amino acids such as serine and arginine, two potentially important amino acids for cell growth and protein production, varies widely between the media tested here while other amino acids are more consistent between media.
COMPACT WITHOUT COMPROMISE
The REBEL delivers high method accuracy and reproducibility, ensuring confidence in results. In a spiking experiment, 17 analytes with known concentrations were added into a chemically defined medium and measured using the REBEL. Values obtained with REBEL showed high accuracy (values were within +/- 20% of expected values) and high analytical precision (%RSDs were between 7 to 16%) as demonstrated in Figure 2.

Optimize Your REBEL’s Performance with a Preventive Maintenance Plan
1. REBEL Autosampler Sample Needle
Routine PM replacement and alignment of the autosampler needle will reduce chances of leaks, clogs, and potential missed injections during analysis.
2. REBEL Autosampler Sample Syringe
The autosampler syringe is responsible for all the fluidic movement and precise sampling within the REBEL. Over time the seals within the syringe can wear resulting in leaks and lowered analysis precision of fluidic transfer/sample loads.
3. REBEL Gas System Inlet Filter*
Annual PM replacement of the air inlet carbon and HEPA filters is to ensure optimal performance for the gas handling system in the REBEL. If not replaced the filters become less efficient and could result in poor analysis performance and contamination of more expensive components

4. REBEL Waste System Filter*
A particulate filter included in the waste system within the REBEL is to prevent material from entering the membranes of the waste pump. Routine replacement prevents clogging and decreased waste removal performance.

5. REBEL MS Detector Inlet*
Detector Inlet is the entrance to the mass spectrometer of the REBEL device. Routine maintenance can reduce the chance of clogging, detection issues, and fouling. During PM, the performance of the vacuum system is evaluated along with alignment and detector performance.

Prior to HJB’s integration of the REBEL™ cell culture media analyzer into their processes, the upstream process development team, led by Vice President Lisa Zheng, sent media samples off to HJB’s analytical sciences group for amino acid analysis. But relying on another team for results meant they were waiting to make decisions and take next steps in their media development processes.
In their search to accelerate R&D and reduce costs, HJB discovered the power of REBEL and ZipChip.
SOLUTION: Answers at-line, anytime
Lisa’s team purchased a REBEL and set it up right next to their bioreactors. They found the device space-friendly and easy to install. Consumables were easy to maintain, and operation was so simple that even their non-technical analysts could conduct analysis. Samples were measured quickly, and with results in less than 10 minutes, scientists were making decisions in near-real time. REBEL is now used routinely for quick analysis to support accelerated media and process improvements.
CONCLUSION: A new milestone in productivity
Because REBEL lets the upstream process development group analyze many samples in a short period of time, the group accumulates a large amount of amino acid data just through daily testing. This lets them quickly investigate deviations during product development, carry out process development/optimization based on REBEL data, and better understand the impacts on culture performance and Product Quality Attributes (PQAs) from process parameter changes.
REBEL was also key in helping HJB accelerate media and process development, which is essential for the development of highly intensified, continuous cell culture perfusion. Earlier this year, the company announced that its continuous perfusion cell culture platform has achieved industry-leading volumetric productivity of greater than 6 g/L per day while maintaining process and product quality attributes in a state of control for a 4-week culture.

CHALLENGE: A fair share of challenges with LC
Liming Shi is the VP in charge of the Analytical Science and Quality Control Departments, and responsible for Product & Process Development Operation. HJB’s Analytical Science Department develops and optimizes analytical methods, and provides analytical support for cell lines upstream, downstream, and formulation process development. The group applies the most advanced analytical techniques to perform large molecule characterization, including analysis of mAbs, ADCs, and bispecific antibodies.
Liming’s team was looking for difficult-to-separate charge variants, which is a challenge with LC-based separations. They were also bumping into UHPLC challenges, such as poor separation and analysis with limited sample volumes. Furthermore, their UHPLC-MS separation and analysis had a turn-around time that was too long.
SOLUTION: In comes ZipChip
In their search for analytical tools that could provide higher-quality data and get them to results faster, the Analytical Services team put ZipChip® straight to work.
Because ZipChip enables rapid measurement of Critical Quality Attributes (CQAs) of native and intact monoclonal antibodies (mAbs), the team used it to characterize intact mAbs and assess the root cause of increases in acidic and basic variants under stress at high temperature. The antibody was analyzed at multiple levels, including its intact state under native conditions and subunit and peptide levels. They used the same ZipChip system and method for both analyses, and were able to determine charge heterogeneity speciation and identification in under 10 minutes. This new workflow was 100 times faster with less prep compared to conventional LC, which required them to harvest fractions.
It was also easy to start integrating ZipChip into their other laboratory workflows as minimal method development was needed for different types of proteins. The team noted this was a big benefit when it came to analysis of native proteins.

They’ve adopted ZipChip into their intact antibody analysis workflows (denatured and native conditions) and their subunit and peptide mapping analysis as it gives them very clear and in-depth information on antibody molecules. One of the stand-out features in Liming’s opinion is having an online platform that provides straightforward separation and identification of charge variants based on different mechanisms.

More specifically, Raman-based optical, in-line measurement methods have shown promise as a way to monitor critical performance parameters, including glucose and lactate concentrations, in real-time. Recently, this has translated to the implementation of Raman-based glucose feeding strategies to modulate productivity and product quality in Chinese hamster ovary (CHO) cell cultures. The current work implements a novel Raman-based solution that does not require any chemometric models for glucose, lactate and biomass measurements. MAVERICK was directly, without any training in the specific bioprocess, deployed to dynamically control lactate feeding. We compared feeding strategies as means of modulating product quality and improving culture conditions, primarily through decreased ammonia accumulation. In contrast, CAR T-cells become less energetic and more glycolytic, which are known features of T-cell exhaustion. Further, metabolic assessment with multiple methods produced findings that correlated with this assessment, such oxygen consumption and certain amino acid levels in spent media. Finally, CAR T-cell activity, potency, and cytotoxicity assessments showed a correlation with anti-tumor activity. Together, this work demonstrates that distinct metabolic features allow CAR/VIPRa T cells, when antigen-stimulated, to have enhanced functions against tumors. Developed in partnership with Emory University and the Winship Cancer Institute, this poster has made waves at the 2024 ISCT & ISCT EU Conference, the ESACT Conference, and Advanced Therapies 2024.

Overview:
Liquid chromatography-mass spectrometry (LC-MS) is used in various applications across the biotherapeutic protein manufacturing workflow pertaining to drug product quality. Mass spectrometry-based applications range from evaluating glycan distributions in monoclonal antibody products to optimization of the cell culture process development by monitoring amino acid and metabolite consumption. Given its analytical flexibility, LC-MS has the capacity to address complex questions; however, LC-MS instrumentation requires a high level of technical experience and more method development than other analytical procedures. Additionally, media development and feed studies in cell culture process optimization are labor-intensive and time-consuming. There is a growing interest in implementing efficient and simplified methods for cell culture media analysis to push for faster innovative processes in biomanufacturing. Newer analytical equipment does not attempt to address the complexity of LC-MS applications specifically targeted for high throughput media analysis of cell culture. This capillary electrophoresis-mass spectrometry (CE-MS)-based Amino Acid Analyzer is a device with simple procedures that provide a similar ability to the traditional LC-MS to specifically identify and quantitate an array of amino acids and several other important metabolites while providing a user-friendly interface and process.

Despite the remarkable clinical efficacy of chimeric antigen receptor (CAR) T cells in hematological malignancies, only a subset of patients achieves a durable complete response (dCR). DCR has been correlated with CAR T cell products enriched with T cells of memory phenotypes. Therefore, reagents that consistently promote memory phenotypes during the manufacturing of CAR T cell products have the potential to significantly improve clinical outcomes. Researchers at Emory University, led by Dr. Sarwish Rafiq, have developed a novel modular multi-cytokine backpack (MCB) platform that combines the signals necessary for activation, co-stimulation, and cytokine support into a single “all-in-one” reagent for CAR T cell manufacturing.
This platform allows for the assembly and screening of compositionally diverse MCB libraries to identify formulations tailored to promote specific phenotypes with a high degree of flexibility. We leveraged this platform to identify unique MCB formulations that increase the proportion of memory phenotype CAR T cells. MCB-manufactured CAR T cell products exhibit increased proportions of memory-like phenotypes in diffuse large B cell lymphoma (DLBCL) patient CAR T cells and demonstrate superior anti-tumor efficacy in mouse models of lymphoma and ovarian cancer through enhanced persistence.
In addition, we saw indications of differences in the metabolism of the MCB-manufactured T cells through spent media amino acid analysis. This analysis was performed with 908 Devices REBEL at-line analyzer and showed nutrient consumption and metabolite accumulation profiles which may indicate different metabolic pathways being active in memory-like phenotype CAR T-cell products.
Our findings serve as a proof-of-principle of the powerful utility of the MCB platform to identify reagents that can improve the effectiveness of cell therapy products through optimal manufacturing.

Overview:
Liquid chromatography-mass spectrometry (LC-MS) is used in various applications across the biotherapeutic protein manufacturing workflow pertaining to drug product quality. Mass spectrometry-based applications range from evaluating glycan distributions in monoclonal antibody products to optimization of the cell culture process development by monitoring amino acid and metabolite consumption. Given its analytical flexibility, LC-MS has the capacity to address complex questions; however, LC-MS instrumentation requires a high level of technical experience and more method development than other analytical procedures. Additionally, media development and feed studies in cell culture process optimization are labor-intensive and time-consuming. There is a growing interest in implementing efficient and simplified methods for cell culture media analysis to push for faster innovative processes in biomanufacturing. Newer analytical equipment does not attempt to address the complexity of LC-MS applications specifically targeted for high throughput media analysis of cell culture. This capillary electrophoresis-mass spectrometry (CE-MS)-based Amino Acid Analyzer is a device with simple procedures that provide a similar ability to the traditional LC-MS to specifically identify and quantitate an array of amino acids and several other important metabolites while providing a user-friendly interface and process.
