High glucose concentrations can also negatively impact protein quality attributes. To address these challenges, we developed MAVERICK, a novel Raman-based PAT approach that enables real-time monitoring and control of CHO cell cultures without the need for prior model building. This versatile approach can be applied across various cell lines, media systems, reactor scales, and culture modes. By implementing a continuous dynamic feeding strategy guided by MAVERICK, we can enhance process performance and improve protein quality attributes like glycosylation, glycation, and charge variants.
MAVERICK, a novel Raman-based PAT technology, was showcased at the ECI Integrated Continuous Biomanufacturing Conference 2024. This technology allows for immediate monitoring of critical process parameters like glucose, lactate, and biomass without the need for extensive preliminary work or model building. By implementing a continuous glucose feeding strategy guided by MAVERICK, researchers were able to significantly improve process titer, cell-specific productivity, and protein quality attributes. This straightforward approach highlights the potential of MAVERICK to revolutionize biomanufacturing processes.

Current approaches to predicting these parameters rely heavily on latent variable/regularized empirical calibration methods, with partial least-squares (PLS) being the most dominant. Although one advantage of these approaches is that they require zero detailed understanding of the underlying system (physics/chemistry/biology), there are several significant disadvantages to modelling bioprocess under these frameworks. Understanding how bioprocess changes impact product quality is crucial for successful biopharmaceutical development. However, frequent data collection across the entire bioprocess can be a significant challenge for analytical teams.
This webinar explores a streamlined solution for acquiring critical mAb quality attributes, including charge variant profiles, sequence identity, glycosylation profiles, and glycation levels. We will demonstrate a simple "dilute-and-shoot" analysis utilizing ZipChip capillary electrophoresis coupled with high-resolution mass spectrometry.
Furthermore, we will present a case study comparing CHO fed-batch processes with traditional daily bolus glucose feeding and a 2g/L dynamic glucose feed strategy, enabled by MAVEN monitoring and control. This study highlights the clear impact of process feeding strategies on product quality profiles.
Presenters
- Erin Redman, PhD, Principal Scientist, 908 Devices
- Graziella Piras, PhD, Senior Director of Strategic Marketing, Life Science, 908 Devices
Key Takeaways:
- Streamline mAb product quality assessment throughout bioprocess runs.
- Gain insights into charge variant profiles, sequence identity, glycosylation profiles, and glycation levels.
- Understand the link between process feeding strategies and product quality.
- Learn how bioprocess monitoring enables control, such as stabilizing glucose levels at 2g/L via MAVEN control to reduce glycation.
- Explore the benefits of both off-line analytical and on-line process analytical technology (PAT) solutions.

Process intensification requires a thorough understanding of the process to achieve desired yields while improving resource utilization and maintaining consistent product quality. Continuous monitoring of glucose and lactate provides deeper process understanding. Dynamic glucose control can lead to better cell viability, lower levels of lactate production, and therefore, higher titer and better control of quality attributes (CQAs) in fed-batch and intensified processes. Learn how you can make more informed decisions with in-line, on-line and at-line analytics of key process parameters

In this webinar, we discuss the implementation of MAVERICK for continuous CHO feed control, including novel feeding approach. While empirical Raman spectroscopy models are convenient due to their 'black box' properties, they may prove impractical and insufficiently robust for deployment as Process Analytical Technology (PAT) solutions in complex upstream bioprocesses. In contrast, the MAVERICK platform, based on Raman technology with a de novo approach, operates as effortlessly as a pH-meter, providing real-time measurements of key process parameters such as glucose, lactate, and biomass.

Drs. Chris Brown and Graziella Piras provide information about a new and transformative means to instantly implement in-line monitoring for multiple key process parameters, right out of the box.
It’s been nearly 20 years since the FDA and EMA published guidelines on process analytical technologies (PAT) to drive the advancement of Biopharma 4.0, with the ultimate aim of predictive control and Quality by Design. Over the years several multivariate optical sensing technologies have demonstrated tremendous promise for in-line continuous monitoring of a range of parameters. These technologies offer the promise of hands-off, non-destructive, and easy-to-integrate multi-parameter analysis with feedback control loop. Yet, they often require substantial expert configuration and set-up time that can take months or years, and substantial expense. In this webinar, review the current state of several optical technology approaches and their associated strengths and limitations. A new platform that greatly accelerates successful implementation of in-line monitoring of key bioprocess parameters is discussed.
Key Takeaways:
- Current state of state-of-the-art in-line upstream bioprocess analytics
- A new solution overcoming major hurdles of implementation of in-line multivariate bioprocess analysis
