Understanding bioreactor metabolomics and its relation to protein quality attribute are important aspects of the quality by design (QbD) approach for therapeutic proteins, therefore it is necessary to develop analytical methods for small molecule nutrient/metabolites and proteins from the culture media. Because of the different separation resin utilized, chromatographic separation usually requires different methods for measuring small molecules and proteins. As a result, the throughput could be hampered due to the time spent on developing and running two sets or more methods. In this webinar, we will present a novel approach for simultaneously monitoring small molecules and protein in bioreactor culture media using capillary electrophoresis-mass spectrometry (CE-MS). CE has enabled the simultaneous measurement because the background electrolyte is compatible with separating both small molecules and proteins. By using a commercial CE-MS system called ZipChip, we demonstrate the feasibility of monitoring amino acid and protein titer from clarified cell culture media. Register now.
